  Better ways to diagnose malaria at the point-of-care are critically needed.Antibody-based rapid diagnostic tests have contributed to malaria management, but the use of antibodies for molecular recognition holds several drawbacks for point-of-care tests for the developing world.We have been developing DNA aptamers as new tools for molecular recognition of malaria diagnostic targets.Here, we present new DNA aptamers that recognize Plasmodium lactate dehydrogenase and solve the structure of the aptamer-target complex.The complex reveals the mechanism for discriminatory recognition relative to the human lactate dehydrogenase and provides insight into how we may be able to further optimize and improve the aptamer.We also present DNA aptamers that can recognize histidine-rich protein Ⅱ, a specific marker for Plasmodium falciparum infection.We demonstrate application of these DNA aptamers in various assays that may be developed for clinical application at the point-of-care.
