β-Phenylethyl isothiocyanate(PEITC)is one of the best studied members of the isothiocyanates family.Present study explores the key miRNA which plays a vital role in sciatic nerve conditioning injury promoting repair of injured dorsal column and validates its function in cell level.To further research the substitution therapy of sciatic nerve conditioning injury,the substitution effect of PEITC was investigated.Compared with control group,miR-17-5p of sciatic nerve conditioning injury group were downregulated at 3rd,7th and 14th day post dorsal column lesion and may perform a biological function which is different from its upregulation in simple dorsal column lesion group.After inhibiting miR-17-5p in DRG neurons,the expression of GAP-43 was upregulated and neurite growth is extended.TargetScan database predicated that the target gene of miR-17-5p could be STAT3.It has also been reported that p-STAT3 protein can induce GAP-43 protein expression which can further promote axonal regeneration.Present study found that STAT3 and p-STAT3 had opposite trends with miR-17-5p.After inhibition of miR-17-5p,neurite growth is extended and STAT3,p-STAT3 and GAP-43 are upregulated.It has been reported that PEITC can regulated the expression of miR-17-5p.Accordingly our study tries to validate the effect of PEITC on DRG neurons.Among 0,5,10,15 and 20 μM PEITC,DRG neurons exposed in 10 μM PEITC showed the longest neurite length.And further experiments demonstrated that 10 μM PEITC induced the neurite growth via inhibiting miR-17-5p and further upregulating expression of STAT3,p-STAT3 and GAP-43.In conclusion,PEITC may be an effective medicine which can replace the effect of sciatic nerve conditioning injury via miR-17-5p/STAT3/GAP-43 axis.
