Background: Cardiovascular diseases (CVDs) are the major causes of mortality in patients with diabetes.Numerous studies have shown that garlic exhibits diverse biological activities, including antitumorigenesis, antiatherosclerosis, and detoxificatin.In this study, we investigated the effects of garlic oil and its components, diallyl disulfide (DADS) and diallyl trisulfide (DATS), on diabetic cardiomyopathy in rats administered with streptozotocin.Method: Four-week-old weanling male Wistar rats were injected with STZ (65 mg/kg body wt in citrate buffer, pH 4.5) in a lateral tail vein for diabetes induction.Rats received garlic oil (100 mg/kg body wt) DADS (40mg/kg body wt) and DATS(40 mg/kg body wt) by gavage every other day until 3 weeks after the induction of diabetes, and rats treated with corn oil only were use as control.Results: In addition to significantly decreased fraction shortening % (FS%) in diabetic rat hearts determined by echocardiography, increased oxidative marker (3-NY), TUNEL-positive cells, caspase 3, with decreased PI3K-Akt signaling activities were shown in the results.Interestingly, all of these alterations were reversed following both garlic oil and diallyl trisulfide administration in diabetic rats through increasing PI3K-Akt signaling activities and inhibiting apoptosis pathway activities.
