  Purpose Research of c-Met targeted molecular imaging based on targeted peptide Method 1 Construction of c-Met targeted molecular probe.The c-Met targeted peptide was screened from peptide display library,and then we linked to the peptide a linker of HYNIC for chelation of 99mTc to construct c-Met targeted molecular probe.The specific reaction contains the HYNIC-cMBP,Tricine,EDDA and SnCl2 solution,whitin a reaction temperature of 80 ℃ following a cooling time to room temperature.Then the mixture after reaction should be through 18C colum for pure probe.2 Evalution of physicochemical property and cytotoxicity of c-Met targeted molecular probe.Of them,evalution of cytotoxicity was executed by measuring the inhibition of cell proliferation and induction of cell apoptosis,and the logP value reflect lipid solubility of the c-Met targeted molecular probe.3 With three different expression level of cell line,H1993(high level and high amplification),H441(moderate level),H1299(negative),we have measured the cell uptake and cell efflux of c-Met targeted molecular probe.We will certificate the c-Met target of the probe from cell level.4 Then the probe will injected into the nude mice with different tumor models for SPECT imaging.The SPECT imaging experiment were excuted at 0.5,1,2,4h after tail injection Result The c-Met targeted molecular probe was synthesized at a high yields,and H1993 cell line obtained the highest cell uptake value with slowest cell efflux,the highest uptake timepoint is 1h after injection of probe.In contrast,H1299 have a very low cell uptake value at all timepoint.In imaging experiment,SPECT image was clearly visualized in H1993 tumor model,and can not be visualized in H1299.Conclusion The c-Met targeted molecular probe 99mTc-HYNIC-cMBP possess well targeting to c-Met.
