Our previous results indicated that theanine and courmarin have some anticancer activities although the activities are not very strong.In order to enhance the anticancer activity of theanine and develop new anticancer agents, we synthesized new compounds,theanine-courmarin-derivatives (TCD) with theanine and courmarin.Our results have shown that TCD has much stronger activity than either theanine or courmarin on reduction of the viability and migration as well as tumor growth in the tumor-bearing mice.Our results indicated that TCD strongly reduced a serial of protein expressions and/or phosphorylation related to the growth and migration as well as metastasis of the cancer cells.TCD enhanced the expressions of Bax, caspase 3, PARP-I, p53 and p21 proteins and decreased the expressions and/or phosphorylation of Bcl-2, vascular endothelial growth factor receptor (VEGFR), cyclin D, AKT, and NF-κB.We further confirmed that inhibition of the signaling pathway of VEGFR-PI3K/Akt-NF-κB is the important molecular mechanism of action of TCD against the cancer cell growth and migration.Our results suggest that TCD may have the wide therapeutic and/or adjuvant therapeutic application in the treatment cancer.
