高脂饮食对小鼠肾脏离子通道活性影响初探

  目的:代谢综合征主要表现为高血压、高血糖、高血脂和肥胖,然而目前关于代谢综合症患者血压升高的具体机制尚不明确.我们利用高脂饲料喂养FVB小鼠,制造代谢综合征模型,以肾脏中调节离子稳态的Na+-K+-ATP酶(Na+-K+-ATPase)、Na+/H+交换体3(NHE3)、70pS+钾通道(ROMK)、Na+-K+-2Cl-共转运体2(NKCC2)和钠氯共转运子(NCC)为研究对象,初步探讨引起代谢综合征患者血压升高的分子机制.方法:选取20-24周龄的FVB雄性小鼠,分为两组,分别用脂肪含量为25%的高脂饲料和脂肪含量为4%的正常饲料喂养2周.无创鼠尾动脉tail-cuff法结合鼠尾血压仪检测小鼠血压,分别用Realtime-PCR和Western Blot方法检测小鼠肾脏Na+-K+-ATPase、NHE3、ROMK、NKCC2和NCC的表达水平.结果:高脂饲料喂养两周后,小鼠血压变化不明显.Realtime-PCR结果显示5种离子通道的mRNA表达均明显增加. Western Blot结果显示除NCC蛋白表达明显上调,与Realtime-PCR结果一致外,Na+-K+-ATPase、NHE3和ROMK这三种蛋白表达无明显差异,而NKCC2蛋白表达则明显减少,与Realtime-PCR检测结果不一致,说明高脂饮食对NKCC2存在转录后调控.结论:高脂饮食分别在转录水平上调肾脏NCC表达,在转录后水平下调肾脏NKCC2表达,参与代谢综合征的血压调节.
