Transient receptor potential (TRP) channels are polymodal cation channels that provide one of the main pathways for cation influx in excitable and non-excitable cells.Among them, thermo-sensitive TRPV1 and TRPV2 channels are molecular sensors for high temperatures important for physiology and diseases.These channels are nonselective cation channels and permeate Ca2+ on the plasma membrane.The Ca2+ influx through these channels may play major roles in physiological functions, such as regulation of Ca2+ homeostasis and transmitter release.Thus, it is important to determine fractional contribution of Ca2+ to the total ion influx, or their fractional Ca2+ current (Pf) by combing patch-clamp (total current) and fura2 (Ca2+ influx) [1, 2].In the present work, by determining Pf of pore-site-mutations systematically, we have studied structure-function of Pf(TRPV1) and Pf(TRPV2) in HEK293 cells.We found, that Pf of TRPV2 is 5 times of TRPV1 (22 % vs.5.4%).A single mutation can switch their Pf each other.Finally, physiological impact of Pf(TRP) was examined in native DRG neurons.
