  Asymmetric cell division(ACD)is a fundamental process in stem cell self-renewal and differentiation.During the ACD of Drosophila neuroblasts(NBs),the asymmetric localization of cell-fate determinants ensure the proper segregation of fate determinants between daughter cells,thereby endowing them with distinct cell fates.The polarized distribution of Numb is regulated by several factors,including the Par3-Par6-aPKC complex,adaptor protein Partner of Numb(Pon),and the cellcycle kinases Aurora A(AurA),Cdk1/Cdc2,and Polo.Here,we find that Cdk1 phosphorylates Pon at Thr63,thus creating a docking site for the Polo-box domain(PBD)of Polo-like kinase 1(Plk1),andphospho-Pon binding-induced PBD dimerization relieves the autoinhibition of Plk1.Moreover,we demonstrate that the priming Cdk1 phosphorylation of Pon is important for sequential Plk1 phosphorylation.
