Asymmetric cell division plays fundamental roles in generating cell diversity during development,which require the elaborate interaction between extrinsic cues and intrinsic cues.However,the precise nature of this type of interaction and its involving signaling mechanisms that regulate asymmetric cell division are poorly understood.Here we demonstrate that Gsα is expressed in the germinal zone of neocortical neuroepithelium,concomitant with cortical neurogenesis,and dynamically co-localized with the intrinsic polarity cues Par6,aPKC,and Numb in dividing apical progenitors.Targeted ablation of Gsα in the neocortical progenitors causes a striking shift from asymmetric cell division to symmetric cell division by depolarizing Rapl,Cdc42,Par6,aPKC,and eliminating the basolaterally localized Numb.Our results reveal that Gsα mediates a signaling pathway that controls asymmetric cell division by directing the compartmental positioning of polarity cues.
