Functions of Circadian Clock in Bipolar Disorder

  Bipolar disorder is a kind of mood disorder,which has affected 4% population worldwide.As the aetiology and pathogenesis of this disease is ambiguous,60% of patients with bipolar disorder are misdiagnosed as unipolar depression.Besides,49% of patients relapse within two years even with treatment.Thus,the study of biomarkers and pathogenesis of bipolar disorder is of great importance.Till now,numerous studies have shown that rhythms in activity,sleep,blood pressure and hormone secretion are abnormal in bipolar patients.In addition,mood stabilizers such as lithium can increase the amplitude of clock gene expression rhythms.In this study,we took Clockd19 mutant mice as an animal model to investigate the biomarkers and pathogenesis of bipolar disorder.After confirming the anxiety and depression state of mice,we analyzed serum extracts from different time points of mice by liquid chromatography/quadrupole time-of-flight mass spectrometry at both positive and negative ion modes.Statistical analysis indicated the compounds with different rhythmicity between each group.Then we confirmed these compounds by target MS/MS and online databases.This work intends to find new biomarkers and therapeutic targets of bipolar disorder.Besides,its also supposed to provide assistance for the clinical diagnosis and treatment of this disease.
