  Background: The research on protein folding is the frontier of life science and fold classification is the foundation of protein in folding study.Nowadays, protein fold classification relies on experts and different database has different classification standards, so we urgently need to establish a unified principle method for protein folding type classification and a classification database.Based on LIFCA, this paper presents a new scoring function for β-sheet classification.Methods: The first one is the stablishment of the β-sheet template database.We select 31 kinds fold type of β-sheet from LIFCA whose number of samples greater than two.Through the analysis of structure features, we determine the templates of 31 kinds fold type and extract the corresponding characteristic parameters of these templates from DSSP database.The second one is the establishment and evaluation of PH domain-like barrel folding classification.Based on the classification of SCOP1.75, 61 proteins (<25 % sequence identity) of PH domain-like barrel are selected as experimental set.Using the contrast information between the template and the spatial structure of the test proteins, this paper presents a new scoring function Fscore.Establishes a protein folding type classification method based on Fscore and applies it to this folding type classification.The third one is different checking of β-sheet fold type and software implementation.LIFCA database contains 574 samples of 31 kinds β-sheet fold type.We give the result of Sensitivity and Specificity using Fscore and template information of 31 kinds β-sheet fold type.Meanwhile, we give the results of the TMscore as a comparison.Results: The fold type classification mentioned above can be automatically accomplished by the LIFCA-Fscore program.Conclusions: The β-sheet fold type classification method can not only guide research of otherfold types but also lay the foundation for automatic fold type classification .
