Objectives: Clongning the gene of AnMCSU from Ammopiptanthus nanus.The function of this gene was verified by Agrobacterium mediated Arabidopsis thaliana.In order to enrich the national genetic resources with the function of anti adversity.It is hoped that this gene is transferred into maize by transgenic method,which improves the ability of maize resistance to reverse.Methods: Clongning the gene of AnMCSU from Ammopiptanthus nanus use RACE method.Using qRT-PCR to find the expression of AnMCSU in heat,dehydration,high salt stress,and ABA induction,and inhibited by cold stress.Build the carrier of pC2300-eGFP to find the function genes in where.Build the carrier of pC2300-35S-AnMCSU transformation Arabidopsis to known the fuction of AnMCSU.Results: The full-length cDNA of AnMCSU is 2544bp long and encoding an 84.85kDa protein.The result of qRT-PCR Showed that the expression of the AnMCSU gene was induced by heat,dehydration,high salt stress,and ABA induction,and inhibited by cold stress.Subcellular localization shown that the protein is located in the cytoplasm.Conclusion: The full-length cDNA of the molybdenum cofactor sulfurase geneAnMCSU from A.nanus is 2544 bp long and contains an open reading frame of 2289 bp,encoding an 84.85 kDa protein of 762 amino acids.Expression of the AnMCSU gene is strongly induced by heat,dehydration,high salt stresses,and ABA induction,and inhibited by cold stress.Its encoded protein is located in the cytoplasm.Theheterologous expression of the AnMCSU gene confers high salt,cold,anddehydration tolerance to Arabidopsis.
