Chlorogenic acid enhances intestinal barrier of weaned rats through tight junction proteins

  Chlorogenic acid,a natural phenolic acid present in fruits and plants,provides beneficial effects for human health.The obj ectives of this study were to investigate whether chlorogenic acid (CHA) could improve the intestinal barrier integrity for weaned rats with lipopolysaccharide (LPS) challenge.Thirty-two weaned male Sprague Dawley rats (21±1d of age; 62.26±2.73g) were selected and randomly allotted to four treatments,including weaned rat control,LPS-challenged and chlorogenic acid (CHA) supplemented group (orally 20mg/kg and 50mg/kg body).The LPS as the stress-inducing agent was injected on d 13 0f the 14 days experiment.CHA supplementation significantly decreased the concentration of serum BUN and ALB compared to LPS-challenged group (P<0.05).It also reduced the level of IFN1 and TNF-a in the jejunal and colon of weaned rats (P<0.05).CHA administration increased villus height and the ratio of villus height to crypt depth in jejunal and ileal mucosa by LPS challenge (P<0.05).CHA supplementation decreased intestinal permeability indicated by the ratio of lactulose to mannitol and serum DAO activity compared to weaned rats with LPS challenge.Immunohistochemistry analysis of tight junction proteins revealed that ZO-1 and occludin protein expression in jejunal and colon were increased by CHA (P<0.05).In addition,immunoblot analysis revealed that occludin level in colon were increased by CHA (P<0.05).In conclusion,CHA enhanced the intestinal barrier function and decreased the intestinal permeability in LPS-challenged weaned rats through increasing tight junction proteins.
