强直性脊柱炎异位骨化机制的研究进展

  强直性脊柱炎(Ankylosing Spondylitis,AS)是一主要侵犯骶髂关节、脊椎关节与附近的肌腱、韧带等软组织,炎症后继发纤维化与异位骨化,使脊椎逐渐失去柔软度,严重时会发展到像“竹节”一样无法弯曲或伸展.在目前已经发现的与强直性脊柱炎有关的多个易感基因中,与AS异位骨化关系最密切的是BMP基因,其他AS易感基因如ARTS1,ILR23可能也与骨化有关,但还需进一步研究,AS骨化易感基因的研究为从分子遗传学的角度发现强直性脊柱炎的骨化机制提供了线索.AS骨化的主要病理特征是起止点炎(enthesitis)以及炎症后的纤维化与骨化,形成骨赘和“竹节样”病变.

根据NCBI提交的刺参甘露糖结合凝集素(Mannan-binding lectins,MBLs)序列(AY625513)设计引物,抽取刺参体腔液进行总RNA提取,采用RT-PCR技术,获得498bp目的片段。通过克隆、测序确定得到刺参MBLs序列。将得到序列进行核苷酸和氨基酸序列比对,首次鉴定了大连刺参MBLs序列,并为我国不同区域刺参的进一步分类提供了科学的分子水平依据。

目的:通过转染hTERT基因构建强直性脊柱炎成纤维细胞永生化细胞系并观察期转化特征。方法:取强直患者韧带组织通过胶原酶消化法分离培养原代成纤维细胞。细胞稳定传代后转染PCI-NEO-hTERT质粒,经G418筛选得到阳性克隆,连续传达培养。应用PCR-ELISA法检测细胞端粒酶活性。采用细胞形态学观察、染色体核型分析、免疫荧光分析转染细胞是否具有转化特征。结果:转染后获得阳性克隆扩大培养和生长良好
