Chemical approaches coupled with mass spectrometry have been widely employed for functional and quantitative proteomic studies.In this talk, we will present our recent results on the use of stable isotope labeling by amino acid in cell culture (SILAC) along with a membrane-impermeable lysine-reactive probe for assessing the differential expression of surface membrane proteins on primary and metastatic tumor cells.
