二甲基甲酰胺对大鼠肝脏线粒体生理功能的影响

  二甲基甲酰胺(DMF,分子式C3H7NO)是一种用途很广的有机溶剂,亦用作萃取剂、医药和农药杀虫脒的原料.浓度较高的DMF 可能引起肝、肾及胃的损坏.线粒体是细胞中重要的细胞器.在研究药物与线粒体的相互作用时,也常用DMF 作为助溶剂,DMF 可能会对线粒体的生理功能产生影响.因此,我们研究了DMF 对Wistar 大鼠肝脏离体线粒体功能的影响.运用紫外-可见吸收光谱、荧光光谱等方法,测试了DMF 对离体线粒体的膜肿胀、氢离子渗透性、钾离子渗透性、膜电势、膜流动性等生理功能的影响.实验结果表明:低浓度的DMF 对线粒体功能影响不大,随着加入体积比的升高,线粒体肿胀程度增大(Fig.a),膜流动性增强(Fig.b).
