Our laboratory has demonstrated a critical role for autophagy in breast cancer cell survival and resistance to antiestrogen therapy [Mol Cancer Ther 2008:7:2977-87].We have demonstrated that 3-methyladenine or bafilomycin, two inhibitors of autophagy formation/function, significantly enhance the cytotoxic action of antiestrogen therapy in cultured breast cancer cells.In addition, we have uniquely identified the proteasome inhibitor bortezomib to effectively block autophagic protein catabolism, in addition to its already known function of blocking proteasome-mediated turnover of proteins.
