哺乳动物基因组DNA 上胞嘧啶第五位碳原子的甲基化修饰(5mC)是一种非常重要的表观遗传调控方式,在基因印迹、X染色体失活、转座子与外源DNA的沉默及组织特异性基因的表达调控中发挥着重要的作用。近年来,研究者发现Tet双加氧酶家族蛋白能够催化5mC氧化生成5-羟甲基胞嘧啶(5hmC),并可以进一步将5hmC氧化成5-甲酰胞嘧啶(5fC)和5-羧基胞嘧啶(5caC),然后糖苷酶TDG能够特异性地识别并切除DNA中的5caC和5fC,进而启动碱基切除修复途径(BER)完成DNA去甲基化。
