Biogenic amines(BAs)are nitrogenous compounds with low molecular weight,including histamine,putrescine,cadaverine,spermine,spermidine,tyramine,tryptamine,phenethylamine and agmatine,etc.BAs are mainly from decarboxylation of amino acids and usually exist in the foods containing high level of protein and amino acids,such as aquatic product,meat,wine.When the foods are metamorphic,the amount of BAs will increase and it will be harmful to human body,causing allergies,headaches,breathing difficulties and some other bad symptoms.Therefore,BAs can be used as indicators of foods' spoilage to avoid the disease caused by BAs.At present,there are many analytical methods for BAs detection,such as gas chromatography(GC),capillary electrophoresis(CE),high-performance liquid chromatography(HPLC),combining with ultraviolet(UV),fluorescence(FD)and mass spectrometric(MS)detection.In our experiment HPLC-MS/MS operated in the multiple reaction monitoring(MRM)mode was performed for the analysis of BAs.Due to BAs' polarity,derivatization needs to be adopted for the reversed phase liquid chromatography(RPLC)analysis.Benzoyl-Cl was selected as the derivatization reagent for nine BAs.For sample pretreatment,sulfosalicylic acid was used as the extraction reagent of BAs.It has less influence on MS instrument than trichloroacetic acid and perchloric acid described in literature.What's more,employing sample pretreatment method with less sample and extract reagent was more environmentally friendly.The established method was validated with a linearity over the 0.05~500 ng/ml for nine BAs and the correlation coefficient(R2)over 0.996.The lowest detection limit was 1 pg/ml.The inter-day RSD of eight BAs(except phenethylamine with 17.53%)was below 10%,implying good precision.Satisfactory recovery was obtained from 83.8%~113.64%.The stability of the derivatives was investigated by the RSD of peak area.The RSD for most BAs within 48 h was below 15%.The results suggested that stability and precision of our method were acceptable for the analysis of BAs.This method was further applied for the analysis of BAs level in different fishes.
