  To explore the development of a vaccine against transmissible gastroenteritis virus (TGEV) infections,the gene coding for the TGEV protein(S) was inserted into the swinepox virus (SPV).We obtained a recombinant SPV (rSPV-SA) that expresses a TGEV protectlive antigen and that acts as a potential vaccine.Results of PCR and indirect immunofluorecence detection indicated the construction of recombinant virus was successful.For in vivo experiments,immune responses of the vaccination vector were evaluated in the mouse model.The Indirect ELISA assay suggested that when mice were vaccinated with rSPV-SA,the IgG against TGEV were dramatically enhanced particularly during the second inoculation the rSPV-SA.The Cytokine assay show that both Thl-type and Th2-type cytokine levels increased dramatically after vaccinated the rSPV-SA.The neutralization assays results indicate that serum from BALB/C mice vaccinated with rSPV-SA effectively neutralized the CPE production by TGEV.We conclude from our data that our novel recombinant SPV construct could be a potential vaccine against TGEV infection.
