Comparison of different SSR detection techniques and their application in genetic diversity analysis

The PCR products of 85 peanut cultivars, amplified with nine pairs of SSR primers, were detected by silver-staining method and fluorescence detection system, respectively.Using two types of methods, 33 and 69 allelic variations were detectable respectively, which is 3.56 and 7.78 allelic locus on average per primer.Therefore, compared with the results detected by silver-staining method, the fluorescence detection system has the following advantages such as high sensitiveness and high efficiency, and the results obtained by this method is more accurate.
