Ptdlns(3)P regulates autophagy, at least partially, by recruiting WD40 repeat Ptdlns(3)P-binding effectors to autophagic structures.The WD40 repeat Ptdlns(3)P effector Atg18 forms a complex with Atg2 and is recruited to the PAS in yeast via its binding to Ptdlns(3)P.Mammalian Ptdlns(3)P effectors include the WIPI ((W)D40 repeat protein interacting with phosphoinositides)family proteins, which have been classified into two large paralog groups, WIPI1/2 and WIPI3/4.Here we generated mice deficient in Wipi3 and Wipi4 to investigate their role in the autophagy pathway.
