Restoring hippocampal metaplasticity by rapid antidepressant ketamine

  Recent studies indicate that ketamine, a noncompetitive NMDA receptor antagonist, can produce rapid antidepressant effects within hours that sustains for weeks in patients resistant to conventional antidepressants that increase central serotonergic (5-HT) neurotransmission.We ask whether and how ketamine modulates synaptic plasticity in the schaffer-CA1 pathway of the hippocampus in rodents that is known to be involved in the pathology of major depressive disorder (MDD).Using in vivo field potential recordings, we found that metaplasticity at schaffer-CA1 pathway of the hippocampus is disrupted in forced swimming model of depression.However, after ketamine injection, metaplasticity in the same group of animals is restored to the level comparable to naive animals, apparently through mTOR-dependent synaptic formation.Therefore, the rapid antidepressant action ofketamine may be mediated by restoring metaplasticity at schaffer-CA 1 pathway of the hippocampus in rodents.
