  To construct and identify the the plasmid containing Human Argonaute2 coding gene and express it in HeLa cell line.Methods: The total RNA was extracted from HeLa cells.The cDNA encoding human full-length Argonaute2 (AGO2)gene was obtained by RT-PCR.As demonstrated by electrophoresis and sequencing,the gene was subcloned to Fluorescent Protein Expression Vector pKaded-MN1 to construct recombinant expression vector pKaded-MN1/AGO2, then transfected the recombinant into HeLa cells.Results: Restriction endonuclease digestion analysis,sequencing and fluorescence imaging showed that the target gene Argonaute2 was cloned into pKaded-MN1.Expression of human Argonaute2 gene in the transfected HeLa cell was identified with RT-PCR.Conclusions :The recombinant plasmid containing Human Argonaute2 gene has been achieved successfully and then expressed in HeLa cells.
