Pituitary adenoma (PA),tumor occurring in the pituitary gland,is one of the most common intracranial adenomas.Although PA is usually benign and non-metastatic,it can result in serious disorders such as infertility, hypertension and diabetes mellitus.35% of PA can invade the dura mater, cranial bone and sphenoid sinus, which may lead to cerebrospinal or systemic metastasis (pituitary carcinoma).Changes in human endocrine function and metabolism were observed in PAs tumorigenesis, but its pathogenetic mechanism remains elusive. In this study,a transomic research was conducted to understand PAs pathogenesis from transcriptome and metabolome perspectives.Firstly,a comprehensive meta-analysis on 7 microarray datasets was performed to identify PA-differentiating genes, and 62 genes were consistently discovered from 20 random sampling datasets using a novel consensus feature selection method developed in our lab. Some of those 62 genes were reported as differentially expressed in other experimental studies.Secondly,303 plasma samples (126 of patients and 177 of healthy individuals) were collected from the XINQIAO hospital of the third military medical university.Based on the metabolomic profiling of all 303 samples, our consensus feature selection method identified 86 metabolite markers of statistically differential concentration between case and control. Some markers identified here were repeatedly discovered by previous experimental studies.Thirdly, transomic study was performed by enriching 86 metabolites in KEGG pathway and 62 genes in both pathway and regulatory transcription factor.
