Identification and Biological Characteristics of Purpureocillium lilacinum Strain PLHN

  One parasitic fungus strain PLHN was isolated from female of Meloidogyne sp.,which damaged to eggplant in Hainan province.Cultural and morphological characteristics of PLHN were examined,its rDNA ITS gene and serine protease gene sequence were cloned and analyzed,and its biological characteristics were determined.The results showed as follows: the color of strain PLHN colony on potato dextrose ager (PDA) plate was pale purple,and powdery conidia were produced abundantly on the colony.The back of colony was light beige.Phialides developed at the end of the conidiophore were in whorls of 2 ~ 6 and in size of (2.81 ± 0.43) μm × (6.27 ± 1.18) μm;Conidia were subrotund,colorless and in size of (2.60± 0.40) μm × (2.63 ± 0.38) μm.The sequences of rDNA ITS and serine protease gene of strain PLHN shared 99% ~ 100% similarity with corresponding sequences of some Purpureocillium lilacinum stranis (primitive named Paecilomyces lilacinus) deposited in GenBank.According to its morphological characteristics and molecular biology,strain PLHN was identified as P.lilacinum.The optimal medium was potato dextrose agar (PDA).The optimal pH and temperature for mycelia growth,sporulation and conidial germination were 7.0 and 28℃ respectively.The lethal temperature to conidium was 55℃.And darkness was beneficial to PLHN.
