Cloning and Function Analysis of WRKY2 Gene from Moso Bamboo

  WRKY proteins are involved in biotic and abiotic stress responses.Although the WRKY transcription factor family is well studied in Arabidopsis, it is still largely unknown in moso bamboo.In this study, a PheWRKY2 gene (GenBank Accession Number FP101056.1) was cloned by RT-PCR.The full-length sequence of PheWRKY2 gene was 777 bp, with an open reading frame (ORF) of 531 bp encoding 176 amino acids, having a calculated molecular weight of 20.365 kDa and a predicted isoelectric point of 9.13.PheWRKY2 encoded a peptide harboring only one WRKY domain and one zinc finger, falling into Ⅱ subgroup of WRKY proteins.Its homologous gene, AtWRKY75, was found to be inducible by Pi stress.The coding region of PheWRKY2 gene was constructed into a plant expression vector PCAMBIA2300, and transformed into Arabidopsis thaliana.Total RNA samples were isolated from moso bamboo blades after low Pi stress and quantification RT-PCR were analyzed.The following study will be concentrated on whether PheWRKY2 gene was involved in Pi starvation.This study provides some basic dates for exploring the function of PheWRKY2 gene.
