Inflammation, proliferation, and tissue remodeling are essential to wound healing.The hypoxia microenvironment of the wound promotes cell migration through a hypoxia-heat shock protein 90 alpha (hsp90α)-low density lipoprotein receptor-related protein-1 (LRP-1)_autocrine loop.To elucidate a role for this autocrine loop in burn wound healing, we investigated the expression profile of hsp90αaround the edges of the burn wound and discovered a transient increase in mRNA and protein levels.Experiments performed on the skin cell line HaCaT confirmed these conclusions.The hsp90α inhibitor, 17-dimethyl aminoethylamino-17demethoxygeldanamycin hydrochloride (17-DMAG), was used to assess of the function of hsp90α.Identically, topical application of hsp90α during the early phase of secondary burn wound led to reduced inflammation and increased tissue granulation, with concomitant reduction in the size of the wound at each time-point tested (p < 0.05).Consequently, the epidermal cells at the wound margins progressed more rapidly with an expedited healing process.In conclusion, these results provide a rationale for the therapeutic use of hsp90α in burn wound healing treatment.
