目的通过EGFR特异性单链抗体介导siRNA内化入TKIs耐药肿瘤细胞,恢复药物敏感性.方法 1、单链抗体的设计及纯化:设计EGFR特异性scFv/s-9R核酸表达序列并全序列化学合成后连接于表达载体,经酶切鉴定及测序后转化大肠杆菌BL21(DE3).将scFv、s-9R的单克隆工程菌于37℃培养至OD600约为0.4-0.6时,添加异丙基-β-D-硫代半乳糖苷(IPTG)诱导表达.
