目的观察青黛及其有效成分靛玉红对溃疡性结肠炎(UC)小鼠脾脏CD4CD25Treg细胞Foxp3表达的影响,探讨其治疗UC的作用机制和靶点。方法将50只BABL/c小鼠随机分为正常组、模型组、靛玉红组、青黛组和5-ASA组。除正常组外,其余小鼠采用自由饮用5%葡聚糖硫酸钠(DSS)溶液5天,制备DSS诱导的UC小鼠模型。于造模后第5天开始,予各组小鼠给药治疗7天。观察各组小鼠一般状况及结肠组织病理学改变,同时检测脾脏CD4CD25Treg细胞Foxp3的表达变化。结果与正常组比较,模型组小鼠CD4CD25Treg细胞Foxp3的表达显著减少(P<0.01);各治疗组CD4CD25Treg细胞Foxp3的表达较模型组升高(P<0.01)。结论青黛及其有效成分靛玉红可通过上调CD4CD25Treg细胞Foxp3表达以防治UC。
Objective To observe the effect of indigo naturalis and its active ingredient indirubin on Foxp3 expression in spleen CD4CD25Treg cells of ulcerative colitis (UC) mice and to explore the mechanism and target of its treatment in UC. Methods 50 BABL / c mice were randomly divided into normal group, model group, indirubin group, indigo group and 5-ASA group. Except for the normal group, the remaining mice were treated with 5% dextran sodium sulfate (DSS) for 5 days to prepare a DSS-induced UC mouse model. On the 5th day after the model was established, the mice in each group were treated for 7 days. The general condition and histopathological changes of the mice in each group were observed. At the same time, Foxp3 expression in CD4CD25Treg cells was detected. Results Compared with the normal group, the expression of Foxp3 in CD4CD25 Treg cells was significantly decreased in model group (P <0.01). The Foxp3 expression in CD4 CD25 Treg cells in each treatment group was higher than that in model group (P <0.01). Conclusion Indigo naturalis and its active ingredient indirubin can prevent and treat UC by up-regulating Foxp3 expression in CD4 CD25 Treg cells.
