In preclinical studies during 2002-2003 with intra-cerebral implanted N29 tumours in Fischer-344 rats, we found that a single radiation dose of 6 Gy to the brain dramatically enhances the effect of syngeneic tumour cell vaccine, resulting in 75 % complete remissions.Several other researchers around the world have in preclinical studies also recorded the cooperative effect of a single radiation dose of 6-10 Gy with various immune therapeutic regimes.This presentation summarizes the results of our studies in Lund, and from researchers recording the enhanced therapeutic response of malignant tumours to various combinations of immunotherapy (IMU) with single fraction or hypo-fractionated radiation therapy.Clinical trials of combining immune therapy and conventional fractionated radiation therapy, however, have reported sparse effects.Up to now no clinical studies of combining established immune therapy regimes with a single 8 Gy fraction RT has been reported although the promising preclinical results of such a combination.
