miR-324-5p在先天性心脏病患者血浆中的表达

  先天性心脏病(Congenital Heart Disease,CHD)是一种常见的出生缺陷,主要是由于胚胎期心血管系统发育异常所致.微小RNA (microRNA)是一类内源性的、非编码的小分子RNA,在心血管系统发育及疾病发生过程中发挥中枢调节作用.本课题组前期发现转录因子GLI1在先天性心脏病患者中表达异常,生物信息学分析表明在GLI1基因的3UTR上有miR-324-5p的作用靶点.本研究旨在探讨miR-324-5p在先天性心脏病患者中是否存在表达异常,进而影响转录因子GLI1在先天性心脏病患者中表达.目的 探讨miR-324-5p在先天性心脏病患者血浆中的表达情况.方法 选取2012年4月-2012年12月,来自沈阳军区总医院心血管研究所的先天性心脏病患者血浆标本52例(包括室间隔缺损17例,房间隔缺损29例,法洛氏四联症4例,动脉导管未闭2例)和正常对照9例,提取血浆microRNA,经特异性茎环反转录后采用Real Time PCR检测血浆中miR-324-5p的含量.结果 与正常对照相比,先天性心脏病患者中miR-324-5p的表达量明显降低,p<0.05.结论miR-324-5p在先天性心脏病患者血浆中表达降低,可能影响转录因子GLI1在先天性心脏病患者中表达,导致先天性心脏病的发生.
