Catumaxomab is a trifunctional,bispecific antibody targeting EpCAM and CD3 which redirects T-cells to EpCAM expressing tumor cells and is able to evoke T-cell cytotoxic responses.The hybrid Fc region of catumaxomab provides a third functional binding site,which is able to bind and activate Fcγ receptor (FcγR)-positive accessory cells.Basis of clinical development for catumaxomab is the intraperitoneal treatment of malignant ascites (MA),a typical late-stage manifestation of cancer,associated with a poor prognosis and survival.Major cause of MA is the intraperitoneal spread of tumor cells,leading to abnormal accumulation of fluid within the peritoneal cavity.Most of the epithelial tumor cells are EpCAM positive whereas the peritoneum itself is EpCAM negative.So far paracentesis associated with potential complication is the most common symptomatic treatment.
