  In clinical proteomics study, serum and plasma are frequently used for the detection of early diagnostic biomarkers.Although mass spectrometry (MS) based proteomic research has played an important role in discovering potential candidate biomarkers, now available methods, such as offline 2-DE MS/MS and on-line 2D-LC-ESI-MS/MS, are still of great challenges due to the complexity of proteome.Therefore, powerful protein prefractionation methods, such as high-abundance removal by antibody techniques or multidimensional LC, are often adopted.To achieve MS based protein identification, protein pretreatment including denaturation, reduction, alkylation, desalting and digestion should be performed, which are not only tedious and time-consuming, but also might suffer from sample loss and contamination (e.g., keratins).
