  Objective To provide proof of the relationship between insulin signal transduction pathway and the mechanism of AD (Alzheimers disease) by observing the coexistence of associated proteins of insulin signal transduction pathway with cholinergic neuron specific choline acetyltransferase (ChAT) in the brain of mice and the changes of the proteins expressed in the brain of AD.Methods (1) The coexistences of associated proteins of insulin signal transduction pathway with ChAT in the CA1 region of hippocampus of mice:immunohistochemistry continuous section,double enzyme staining and double fluorescent staining were applied to investigate the coexistence of InsR (insulin receptor),IRS-1 (insulin receptor substrate-1),IRS-2,PKB (protein kinase B,also named Akt),GSK-3β (glycogen synthatase kinase-3β) and IDE (insulin-degrading enzyme) with ChAT.(2) The changes of expression of associated proteins of insulin signal transduction pathway in the brain of AD:Western blot was used to investigate the changes of expression of InsR,IRS-1,PI3K (phosphatidyl-inositol 3 kinase) and MAPK (mitogen-activated protein kinase).
