The natural product depsipeptides,beauveriolide Ⅰ(1) and Ⅲ(2) were originally isolated from the fungal strain Beauveria sp.FO-6979 during a screening program for inhibitors of lipid droplet accumulation in murine macrophages.Depsipeptides 1 and 2prevent lipid accumulation in peritoneal macrophages via acyl-CoA:cholesterol acyltransferase(ACAT) inhibition and importantly,they are orally active and are being studied comparatively alongside more classic ACAT inhibitors,such as CP-113,818 and DuP-128.Alzheimers disease(AD) is now the most common form of dementia,affecting up to 15 million people worldwide.
