Human amniotic epithelial stem cells promote wound healing by facilitating the migration and prolife

  Wound healing is a highly orchestrated physiological process consisting of a complex interaction of cellular and biochemical events.Human amniotic epithelial stem cells (HAESCs)have been shown to be an attractive resource for wound healing because they are more primitive stem cells.However, the exact effects of amnion-derived stem cells on migration or proliferation of keratinocytes, as well as their potential mechanism, is not fully understood.In this paper, we found that HAESCs accelerated the migration of keratinocytes, and induced a remarkable increase in the activity of phospho-ERK, phospho-JNK and phospho-Akt,and blockade with their specific inhibitors significantly inhibited the HAESCs-CM induced migration.Furthermore, co-culture of keratinocytes with HAESCs up-regulated the expression levels of cell proliferation protein Cyclin D1, Cyclin D3 and Mdm2.In vivo animal experiments showed that HAESCs-CM improved the wound healing, while blockade with ERK, JNK and Akt inhibitors significantly impaired wound healing.Taken together, these results showed, for the first time, that HAESCs promoted wound healing by facilitating the migration and proliferation of keratinocytes possibly via ERK.JNK and Akt signaling pathways, and might be a potential therapy in skin wound healing.
