Objective: To investigate the influence of CO2 pneumoperitoneum at different pressures and different exposure times on the chemokine receptor expression of colorectal carcinoma cells.Methods: We developed an in vitro model for pneumoperitoneum.SW480 colon carcinoma cells were selected and exposed to CO2 pneumoperitoneum at different pressures(6, 9, 12, and 15 mmHg) for 1, 2, and 4 h.The cells were cultivated under the same condition as normal SW480 colon carcinoma cells without CO2 pneumoperitoneum treatment,at 37 ℃ and 5％ CO2.The expression of chemokine receptors CXCR4 and CCR7 were detected by immunocytochemistry and RT-PCR after being cultivated for 0, 24, 48, and 72 h.
