  Science and industry community have long waited methodologies for high throughput (HP) kinetic studies of protein-X interactions (X: proteins, small molecules, aptamers and DNA, etc.).We reported herein the application of imaging total internal reflection ellipsometry (iTIRE) [1] for HP study.Comparative studies among four different methods, namely protein microarray, enzyme linked immunosorbent assay (ELISA), chemiluminescence immunoassay (CLIA), and iTIRE will be presented.We will also comment on three imaging methods, namely imaging surface plasma resonance (iSPR), [2] imaging ellipsometry (iELM) [3] and iTIRE.We believe that the kinetic study of protein-X interaction, drug discovery and other related fields will benefit from this iT1RE instrument in combination with chips of nonfouling, functional polymeric matrix prepared via surface initiated polymerization.
