In order to develop and optimize simple and rapid techniques to identify the structural of tanshinones, crude extracts of Salvia miltiorrhiza bung were investigated by liquid chromatography/UV spectrometry with diode-array detection, liquid chromatography/electrospray ionization mass spectrometry (HPLC/ESI-MS), liquid chromatography/electrospray ionization multi-stage tandem mass spectrometry (HPLC/ESI-MSn) and Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR-MS).The tanshinone constituents of Salvia miltiorrhiza bung were exhaustively investigated by HPLC/PDA/ESI-MS and HPLC/ESI-MSn, which allowed the on-line identification of various tanshinones.Using the accurate masses obtained from FT-ICR-MS and the tandem mass spectrometric (MSn) data from an ion trap instrument with ESI source in positive ion mode, a series of tanshinones are reported and rationalized, three isomers at m/z 279 and a pair of isomer at m/z 281 were well separated and identified, another peak at m/z 281 but with different elemental composition was also differentiated (see Table 1).
