肝素硅-阿霉素抗癌药物系统对鸡胚尿囊膜血管生成的影响

  目的 评价肝素硅阿霉素抗癌药物系统对鸡胚尿囊膜(CAM)新生血管生成的抑制作用,为新型抗癌药物的研发提供实验基础.方法 选取150只鸡胚随机分组,进行两部分实验: (1)药物系统对碱性生长因子(b-FGF)诱导的血管生成的作用:分为对照组(NS),b-FGF组(0.1 μg/胚),药物系统高、中、低剂量组(b-FGF+10μ g HDMSNs/胚、b-FGF+5 μ g HDMSNs/胚、b-FGF+2.5 μ g HDMSNs/胚),肝素硅组(b-FGF+10 μg MSNs-Hp/胚),肝素组(b-FGF+Heparin 10 μg/胚),阿霉素组(b-FGF+1 μ g ADR/胚),介孔硅组(b-FGF +10 μ gMSNs/胚),共9组,每组8只.(2)药物系统对正常血管生成的影响:分为对照组(NS),药物系统高、中、低剂量组(10 μ g/胚、5 μ g/胚、2.5 μg/胚),肝素硅组(10 μ g/胚),肝素组(10 μ g/胚),阿霉素组(1 μ g/胚),介孔硅组(10 μ g/胚),共8组,每组8只.在鸡胚发育到第11天时,进行开窗、加药,放入培养箱里继续孵育3天,数码相机拍照,取加药部位及其周围的尿囊膜,甲醛固定后在BI-2000图像分析系统下进行血管分叉数、血管内径计数及拍照.
