Parkinsons disease was a progressive neurodegenerative disorder characterized by the degeneration of dopaminergic neurons located in substantia nigra pars compacta.To evaluate the neuroprotective effects of triptolide(T10) in acute MPTP mice model, experimental Parkinsonism was established in male C57BL/6 mice by intraperitoneal injections of MPTP (20 mg/kg MPTP-HCL, four times at 2 h interval).T10 was pretreated 7 days before MPTP injection, and was constantly used for 3 days after MPTP injection, and then animals were sacrificed atlst, 3rd, 7th day after MPTP injection.Results showed that T10 prevented the loss of dopaminergic fibrils in striatum and dopaminergic neurons in the substantia nigra, and there was a tendency in preventing the decrease of the content of DA and its metabolite-DOPAC, in the striatum of MPTP mice, and reversing the abnormally increased ratio of (DOPAC+HVA) to DA.Triptolide(T10) has neuroprotective effects in acute MPTP mice model.
