Fragment-based drug design (FBDD) has been proved to be a novel paradigm for small molecule drug discovery in the past decade.It has been established as the principle altemative to traditional methods used for lead discovery in pharmaceutical companies,such as high-throughput screening (HTS) and virtual screening.In Astellas,we have introduced FBDD platform into lead discovery process in various drug discovery projects.Given the increasing demand for data regarding protein-ligand interaction,we acquired a priority access to a new high throughput synchrotron beamline AR-NE3A built in the Photon Factory Advanced Ring at High Energy Accelerator Research Organization in this April.We are now trying to take full advantages of a large number of complex structures and also combine FBDD with other technologies.In this work,we will describe the some aspects about application of FBDD including successful and unsuccessful experiences in Astellas.
