Using local chromatin structure to improve RISPR/Cas9 efficiency in zebrafish

  Although CRISPR/Cas9 system has been successfully applied to zebrafish studies,its efficiencies vary greatly for different gRNAs,even for those targeting on the same genes.As the workload and cost of zebrafish mutant screening is largely depended on the mutation ratio of injected embryos,selecting more effective gRNAs is especially important for zebrafish homozygous mutants construction.Besides the influence of sequence features,local chrornatin structures may have effects on CRISPR/Cas9 efficiency in zebrafish,which is largely unexplored.The only related study in zebrafish did not observe the effects of nucleosome organization on CRISPR/Cas9 efficiency,which is inconsistent with the recent in vitro study.
