Manganese is an essential trace element for biological development and normal function.Excess manganese results in immunosuppression.This study was designed to investigate the effect of excess exposure to manganese on the cytokines in chicken spleen lymphocytes.Lymphocytes were incubated in culture medium in the absence or presence of MnCl2 (2×10-4,4×10-4,6×10-4 and 8×10-4 mmol·L-1) for 12,24,36 and 48 h,respectively.The mRNA levels of IL-2,IL-4,IL-6,IL-12β,IL-17 and IFN-γ were examined by real-time PCR.The results indicated that the mRNA levels of IL-2,IL-4,IL-6,IL-12β and IL-17 decreased with the increase of manganese,but the mRNA level of IFN-γ maintained higher quantity.It demonstrated that excess manganese suppressed immune function,and led to abnormal immune reaction at molecule level in vitro experiment.
