AIM To explore the effects of PNS on the content and activity of αt-secretase in the brains of SAMP8 mice with Alzheimers disease.METHODS SAMP8 were randomly divided into PNS 200 and 100 rag· kg-1, huperzine A 0.3 mg· kg-1 group and control group.All were treated with the same volume of water for 8 weeks.The activity of αt-secretase was assayed by direct immunofluoreseent method.Western blot and real-time PCR were used to detect the content of α-secretase proteins and the expression of α-secretase mRNA including ADAM9, ADAM10 and ADAM17 respectively.RESULTS The relative fluorescence units in PNS 200 and 100 mg·kg-1 groups were higher than that in control group at the concentration of 2 g·L-1.Western blot showed that the level of ADAM9 protein expression in PNS 200 and 100 mg·kg-1 and huperzine A groups was significantly higher than that in control group while the level of ADAM10 protein expression in PNS 200 and 100 mg·kg-1 and huperzineA groups was significantly lower than that in control group.The level of ADAM17 protein in PNS 200 and 100 mg·kg-1 was decreased compared to control group.However, The level of ADAM17 in huperzineA group was higher than that in control group.The expression of ADAM9 mRNA in PNS 200 mg·kg-1group and huperzineA group were significantly higher than that in control group.The expression of ADAM10 in PNS 100 mg·kg-1group was lower than that in control group.CONCLUSION PNS can increase activity of α-secretase in the brain of SAMP8 mouse via increasing the level of ADAM9 protein expression.
