  A capillary electrophoresis (CE) coupled with laser-induced fluorescence (LIF) detection method was developed to detect doxorubicin (DOX) uptake in single K562 cells based on drug dosage and exposure time.The K562 cells were treated with different concentration of DOX (1, 3, 10, 20, 30 and 50 μM) for different exposure time (1, 3, 5, 24 and 48 h), respectively.A minimum of 20 cells were analyzed for each DOX-treated cell population.The marked heterogeneity in DOX uptake was observed and the variation of DOX uptake amount (RSDs %) ranged from 24.0 % to 61.1% within cell populations.The cell-to-cell variation in DOX uptake decreases and tends to be constant with increasing DOX concentration, but does not exhibit regular variation with increasing exposure time.The mean intracellular DOX amount is a linear function of extracellular DOX concentration.
