对氯苯丙氨酸对大鼠海马胞液皮质酮受体数量的影响

本实验以(~3H)皮质酮为配体,建立了去肾上腺大鼠海马胞液皮质酮受体的放射配体结合测定法,并对海马胞液皮质酮受体的结合特性以及对氯苯丙氨酸(pCPA)对皮质酮受体数量的影响进行了研究。测得的皮质酮受体具有可饱和性、有限的结合能力、高亲和力和高特异性的特征。Scatchard分析为一直线,表明测得的皮质酮受体是一个或一组kd值十分接近的受体。侧脑室注射pCPA 1mg4天后,海马5-HT含量明显降低,同时海马胞液皮质酮受体的最大结合客量(Bmax)显著升高,提示海马5-HT能神经元的功能性活动在控制海马胞液皮质酮受体数量的相对恒定中具有重要作用。 In this experiment, (~ 3H) corticosterone was used as a ligand to establish a radioligand binding assay of corticosterone receptor in the hippocampus of adrenal rats. The binding properties of corticosterone receptor to the hippocampus and the activity of p-chlorobenzene The effect of alanine (pCPA) on the number of corticosterone receptors was studied. The measured corticosterone receptor has characteristics of saturability, limited binding capacity, high affinity and high specificity. Scatchard analysis was linear, indicating that the measured corticosterone receptor is one or a group of receptors with very close kd values. After intracerebroventricular injection of pCPA 1 mg for 4 days, the content of 5-HT in the hippocampus was significantly decreased, meanwhile, the maximal binding capacity (Bmax) of corticosterone receptor in hippocampus was significantly increased, suggesting that the functional activity of 5-HT neurons in the hippocampus may play an important role in the hippocampus Cytosolic corticosterone receptor in a relatively constant quantity has an important role.
