The wheat polyphenol oxidase distributed mainly in the cortex of the grain,and its content in the flour is less,but its catalytic oxidation of phenolic substances generated brown or black anthraquinones derivatives could seriously affected the wheat products colour and luster,this was the important factors that caused browning on the surface of wheat products,and has serious influence on the quality of wheat products.The properties of wheat polyphenol oxidase were investigated Two wheat varieties were selected as experimental materials and catechol was used as enzyme substrate.The activity of wheat polyphenol oxidase was determined with a spectrophotometry at 410 nm.The effects of pH,temperature,substrate concentration and enzyme concentration on its activity were studied.The results showed that the optimum temperature and pH of wheat polyphonel oxidase was 40℃ and 6.0,respectiveiy.The substrate concentration and enzyme concentration were proportional to the activity of polyphenol oxidase.Therefore,during wheat products processing,the browning of wheat products could be improved through adjust processing technology.
