蛋白质的翻译后修饰对调节蛋白质的生物活性是至关重要的。理解蛋白质翻译后修饰的过程和机制首先需要对翻译后修饰的种类和程度进行分析。我们尝试发展元素标记策略,结合酶选择性和底物代谢机制,开展了蛋白质翻译后修饰的元素质谱定量分析研究。简要介绍基于元素质谱的生物分析[1,2]后,这里主要向大家汇报1)蛋白质酪氨酸磷酸化的选择性分析[3]和2)细胞表面膜蛋白质和脂类唾液酸糖基化的专一性成像和绝对定量的研究进展。
